Indications/Uses
Treatment of primary hypercholesterolemia (type IIa & IIb hyperlipoproteinemia) caused by elevated LDL cholesterol conc in patients unresponsive to diet or non-drug measures. Reduction of elevated LDL in patients w/ combined hypercholesterolemia & hypertriglyceridemia.
Dosage/Direction for Use
Adult Initial: 20 mg once daily. Adjust dose at 4 wk intervals. Maintenance: 20-80 mg daily. Patients on immunosuppressive therapy should be given ≤20 mg daily.
Administration
Should be taken with food.
Contraindications
Active liver disease or unexplained persistent elevation or serum transaminases. Pregnancy & lactation.
Special Precautions
Persistent elevation of serum transaminases up to 3 times the normal values (discontinue). Perform baseline & periodic liver function tests; & baseline serum creatine kinase levels. Chronic or substantial alcohol consumption or history of liver disease. Women of childbearing potential.
Adverse Reactions
Constipation, abdominal pain, dyspepsia, flatulence, nausea, heartburn, dizziness, headache, hepatitis. Rarely, rhabdomyolysis.
Drug Interactions
Fibrates, cyclosporine, nicotinic acid, macrolides, itraconazole, ketoconazole, coumarin anticoagulants, amiodarone, verapamil, nefazodone.
