Cloxacillin sodium.
Description
Each capsule contains CLOXACILLIN SODIUM equivalent to 250mg of CLOXACILLIN.
Free from dust and powder.
Action
Pharmacology: Cloxacilla, a semi-synthetic penicillin, is Penicillinase stable and active against Gram-positive organisms particularly penicillinase producing Staphylococci, Streptococci and Pneumococci.
Pharmacokinetics: Cloxacilla, is well absorbed: administration of higher doses result in a proportionately higher increase in serum levels. Concurrent administration of Probenecid to patient with normal renal functions causes a higher and prolonged serum level. Cloxacillin is excreted by glomerular filtration and renal tubular secretion.
Indications/Uses
Cloxacilla, is used for infections due to Staphylococci resistant to benzyl-penicillin. It is also used for mixed Streptococcal and Staphylococcal infection when the Staphylococci are penicillin-resistant.
Dosage/Direction for Use
For Adults: 1 capsule (250mg) 6 hourly.
Taken ½ to 1 hour before meals.
In severe infections the doses may be safely increased.
The prescribed course should be completed.
Overdosage
Symptoms: Extended effects of the adverse reactions.
Treatment: Supportive.
If allergic reaction occurs, the drug should be discontinued.
Since there is no specific antidote, treatment of penicillin overdose should be symptomatic and supportive. Haemodialysis may aid in the removal of penicillins from the blood.
Serious anaphylactoid reactions require immediate emergency treatment.
Contraindications
Cloxacilla is not to be used in patients with known hypersensitivity to Penicillin.
Special Precautions
Cloxacilla, should be given with caution to patient with histories of allergy. Renal, hepatic and haematologic system should be evaluated periodically during prolonged therapy. Prolonged use of Cloxacilla may result in overgrowth of non-susceptible organisms including fungi or gram-negative bacteria such as Pseudomonas.
Use In Pregnancy & Lactation
Cloxacilla should be used with caution in neonates, pregnancy and lactation.
Side Effects
As with other penicillins, side effects are usually mild and transitory in nature. This may include skin rashes, nausea, vomiting and diarrhea, fever, joint pains and angioedema.
Others include: Hypersensitivity Reactions: Eosinophilia, pruritus, serum sickness- like reaction with fever, chills and myalgia.
Haematologic Effects: Haemolytic anaemia, transient neutropenia, leukopenia, granulocytopenia, thrombocytopenia, agranulocytosis.
Renal Effects: Acute interstitial nephritis, azotemia.
Hepatic Effects: Intrahepatic cholestasis.
Storage
Store below 25°C, away from direct sunlight.
Shelf-Life: 3 years from the date of manufacture.
