Mirogabalin besilate
Indications/Uses
Peripheral neuropathic pain in adult.
Dosage/Direction for Use
Initially 5 mg bid, then increase dose by 5 mg per dosing w/ at least 1 wk interval up to 15 mg bid. May increase or decrease dose between 10 mg & 15 mg bid. Mild renal impairment (CrCl ≥60 to <90 mL/min) Initially 5 mg bid. Daily dose: 10-30 mg. Min effective dose: 10 mg bid. Recommended dose: 15 mg bid. Moderate renal impairment (CrCl ≥30 to <60 mL/min) Initially 2.5 mg bid. Daily dose: 5-15 mg. Min effective dose: 5 mg bid. Recommended dose: 7.5 mg bid. Severe renal impairment including patients on hemodialysis (CrCl <30 mL/min) Initially 2.5 mg once daily. Daily dose: 2.5-7.5 mg. Min effective dose: 5 mg once daily. Recommended dose: 7.5 mg once daily.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Not for causal therapy for neuropathic pain. Closely monitor & treatment discontinuation or dose reduction should be taken if any abnormalities (eg, dizziness, somnolence & loss of consciousness); early symptoms (eg, general malaise, anorexia) are noted. Potential occurrence of obesity; ophth disorders (eg, amblyopia, abnormal & blurred vision, & diplopia). W/drawal symptoms (eg, insomnia, nausea, diarrhea, decreased appetite) w/ abrupt treatment discontinuation. Measure body wt regularly. Gradually reduce dose. Diagnose & treat underlying disease of pain concurrently. Not to operate potentially dangerous machinery (eg, driving car). Hepatic function disorder (eg, increased AST & ALT). Women of childbearing potential should use effective contraception. Pregnancy & lactation. Childn. Elderly (reduced renal function & have tendency to experience falls).
Adverse Reactions
Somnolence, dizziness. Constipation; oedema, gait disturbance; increased wt & hepatic enzyme.
Drug Interactions
Increased Cmax & AUClast w/ probenecid & cimetidine. Decreased attention & balance-function w/ lorazepam & ethanol.
