Per mL Latanoprost 50 mcg, timolol 5 mg
Indications/Uses
Reduction of IOP in patients w/ open-angle glaucoma or ocular HTN who are insufficiently responsive to topical β-blockers, prostaglandin analogues, or other IOP-reducing agents & in whom combination therapy is appropriate.
Dosage/Direction for Use
Adult (including elderly) 1 drop into affected eye(s) once daily.
Contraindications
Hypersensitivity to latanoprost, timolol or benzalkonium Cl. Reactive airway disease including presence or history of bronchial asthma, severe COPD; sinus bradycardia, 2nd-/3rd-degree AV block, overt cardiac failure or cardiogenic shock.
Special Precautions
Stop treatment if increased iris pigmentation ensues. History of atopy or severe anaphylactic reaction; recurrent herpetic keratitis. Patients w/ CV diseases (eg, CHD, Prinzmetal's angina & cardiac failure) & hypotension; 1st degree heart block; severe peripheral circulatory disturbance/disorder (ie, severe forms of Raynaud's disease or syndrome); mild/moderate COPD; labile diabetes; corneal diseases. Aphakic patients, pseudophakic patients w/ torn posterior lens capsule, or in patients w/ known risk factors for macular oedema. Acute attacks of closed angle glaucoma. Macular oedema including cystoid macular oedema. Avoid in active herpes simplex. May mask signs of hypoglycaemia & hyperthyroidism. May cause eye irritation & known to discolour contact lenses. Contact lenses should be removed prior to application & reinserted after 15 min. Concomitant use w/ systemic β-blockers. Not recommended to use concomitantly w/ 2 local β-blockers or prostaglandins. May affect ability to drive & use machines. Hepatic impairment. Not to be used during pregnancy & lactation. Childn. Elderly (increased sensitivity to timolol).
Adverse Reactions
Increased iris pigmentation. Eye irritation (including stinging, burning & itching) & pain. Dizziness; keratitis, dry eye, corneal erosion; palpitations; chest pain; dyspnoea. Latanoprost: Herpetic keratitis; eyelash & vellus hair changes, punctate epithelial erosions, periorbital oedema, iritis/uveitis, macular & corneal oedema, misdirected eyelashes sometimes resulting in eye irritation, iris cyst, photophobia, periorbital & lid changes resulting in deepening of eyelid sulcus; aggravation of angina in patients w/ preexisting disease, asthma, aggravation of asthma; darkening of palpebral skin; joint & muscle pain. Timolol: Angioedema, urticaria, localised & generalised rash, pruritus, anaphylactic reaction; hypoglycaemia; insomnia, depression, nightmares, memory loss; syncope, CVA, cerebral ischaemia, increased signs & symptoms of myasthenia gravis, parasthesia, headache; signs & symptoms of ocular irritation (burning, stinging, itching, tearing & redness), blepharitis, blurred vision & choroidal detachment following filtration surgery, decreased corneal sensitivity, ptosis, diplopia; tinnitus; bradycardia, oedema, arrhtyhmia, CHF, AV block, cardiac arrest; hypotension, Raynaud's phenomenon, cold hands & feet; bronchospasm, cough; dysgeusia, nausea, dyspepsia, diarrhoea, dry mouth, abdominal pain, vomiting; alopecia, psoriasiform rash or exacerbation of psoriasis, skin rash; myalgia; sexual dysfunction, decreased libido; asthenia/fatigue.
Drug Interactions
Latanoprost: Precipitation w/ thimerosal. Timolol: Possible additive effect on IOP &/or systemic β-adrenergic blockade w/ systemic β-adrenergic blockers. Possible additive effect & production of hypotension &/or marked bradycardia w/ catecholamine-depleting drugs. Additive effects on prolonging AV conduction w/ other CV drugs. Concomitant use w/ oral verapamil & quinidine.
