Short infusion 50-60 mg/kg IV daily for 5 consecutive days. Total dose: 250-300 mg/kg/course. Therapy-resistant cases 80 mg/kg daily for 2-3 consecutive days. Courses should not be repeated earlier than 3-4 wk intervals. Continuous infusion 125-200 mg/kg w/ Uromitexan. Administer Uromitexan for further 12 hr w/o Holoxan.
Patients of reproductive age should use contraceptives during therapy & for the following 6 mth. Unilateral nephrectomies; brain metastases, infection, electrolyte imbalance.
GI upsets; alopecia; reduction in blood count; cystitis; reversible encephalopathy. Immunosuppression. Occasionally, renal, hepatic & cardiac dysfunction in rare cases. Risk of secondary malignancies.
Drug Interactions
Neurotoxicity, hematotoxicity & nephrotoxicity may be increased by prior cisplatin therapy. Increased hypoglycemic effects w/ sulfonylureas. Increased bone marrow depression w/ allopurinol. Disturbances of anticoagulant control of warfarin.
