Contents
Per 625 mg FC tab Amoxicillin 500 mg, clavulanic acid 125 mg. Per 1 g FC tab Amoxicillin 875 mg, clavulanic acid 125 mg. Per 228.5 mg/5 mL susp Amoxicillin 200 mg, clavulanate K 28.5 mg. Per 457 mg/5 mL susp Amoxicillin 400 mg, clavulanic acid 57 mg
Indications/Uses
Short-term treatment of bacterial infections in URTI & lower resp tract including ENT, GUT, abdominal, skin & soft tissue, dental, bone & joint infections.
Dosage/Direction for Use
Tab Adult & childn >12 yr Mild to moderate infections One 375-mg tab tid or one 625-mg tab bid. Severe infections One 625-mg tab tid or one 1-g tab bid. Dental infections One 375-mg tab tid for 5 days or one 625-mg tab bid for 5 days. Moderate renal impairment CrCl 10-30 mL/min One 375-mg tab or one 625-mg tab 12 hrly. Severe renal impairment CrCl <10 mL/min Not more than one 375-mg tab 12 hrly. Susp Mild to moderate infections Childn 7-12 yr 10 mL of 228.5 mg/5 mL bid or 5 mL of 457 mg/5 mL bid, 2-6 yr 5 mL of 228.5 mg/5 mL bid or 2.5 mL of 457 mg/5 mL bid. More serious infections Childn 7-12 yr 10 mL of 457 mg/5 mL bid, 2-6 yr 10 mL of 228.5 mg/5 mL bid or 5 mL of 457 mg/5 mL bid. Childn 2 mth to <2 yr Individualized dose based on body wt. Mild to moderate infections 25/3.6 mg/kg daily. More serious infections 45/6.4 mg/kg daily.
Administration
May be taken with or without food: Best taken at the start of meals for better absorption & to reduce GI discomfort. Tab: Swallow whole, do not chew. May be broken in half & swallowed w/o chewing.
Contraindications
Hypersensitivity to co-amoxiclav or penicillin. Possible cross-sensitivity w/ other β-lactam antibiotics eg, cephalosporins. History of allergy or penicillin-associated jaundice/hepatic dysfunction.
Special Precautions
Not to be used in patients w/ allergic history. Life-threatening allergy. Discontinue if skin rash, flu-like symptoms; fever, vesicle, skin & mucous membrane peeling associated w/ SJS occur. Anaphylactoid reactions. Avoid use if glandular fever is suspected. Severe cholestatic jaundice. Overgrowth of non-susceptible organisms. Monitor hepatic function at regular intervals. Not recommended in concomitant use w/ probenecid. Hepatic dysfunction. Moderate or severe renal impairment. 1st trimester pregnancy. Lactation. Oral susp: Not recommended in very young childn w/ immature renal function & w/ GFR <30 mL/min.
Adverse Reactions
Diarrhoea, indigestion, nausea & vomiting; moderate rise in AST &/or ALT; angioedema, oedema, anaphylaxis serum sickness-like syndrome & hypersensitivity vasculitis. Erythema multiforme, SJS, TEN & exfoliative dermatitis.
Drug Interactions
Prolonged bleeding & prothrombin time w/ anticoagulants. Reduced efficacy of OCs. Increased & prolonged blood levels w/ probenecid. Increased risk of allergic reactions w/ allopurinol.
