Leuprorelin acetate
Indications/Uses
Hormone dependent advanced prostate cancer & high-risk localized & locally advanced hormone dependent prostate cancer in combination w/ radiotherapy.
Dosage/Direction for Use
Adult 22.5-mg inj Single SC inj every 3 mth. 45-mg inj Single SC inj every 6 mth.
Contraindications
Hypersensitivity to leuprorelin acetate & other GnRH agonists. Patients who previously underwent orchiectomy. As sole treatment in prostate cancer patients w/ spinal cord compression or evidence of spinal metastases. Women. Ped patients.
Special Precautions
Transient increased serum conc of testosterone, dihydrotestosterone & acid phosphatase during 1st wk of treatment. Closely monitor patients w/ vertebral &/or brain metastases & urinary tract obstruction during 1st few wk of therapy. Decreased bone density, osteoporosis, long lasting testosterone deficiency, increased age, smoking, alcohol consumption, obesity, insufficient exercise; pituitary apoplexy; hyperglycemia, DM; convulsions w/ or w/o history of predisposing factor; ureteral obstruction, spinal cord compression. Increased risk of developing diabetes, MI, sudden cardiac death & stroke. Monitor signs & symptoms of CV disease. Periodically monitor blood glucose &/or HbA1c, ECG & electrolytes. Concomitant use w/ drugs prolonging QT interval. May affect ability to drive & use machines.
Adverse Reactions
Hot flashes; ecchymoses, erythema; fatigue, inj site burning & paraesthesia. Nasopharyngitis; nausea, diarrhea, gastroenteritis/colitis; pruritus, night sweats; arthralgia, limb pain, myalgia, rigors, weakness; urinary infrequency, difficulty in micturation, dysuria, nocturia, oliguria; breast tenderness, testicular atrophy & pain, infertility, breast hypertrophy, erectile dysfunction, reduced penis size; malaise, inj site pain, bruising & stinging; hematology changes, anaemia; increased blood creatine phosphokinase, prolonged coagulation time.
