Donepezil HCl
Indications/Uses
Dosage/Direction for Use
Initially 5 mg once daily. Mild to moderate Alzheimer disease 5 or 10 mg once daily. Moderate to severe Alzheimer disease 10 or 23 mg once daily. A dose of 10 mg should not be achieved until the patient have been on a daily dose of 5 mg for 4-6 wk. 
Administration
May be taken with or without food: Best taken at bedtime.
Contraindications
Hypersensitivity to piperidine derivatives.
Special Precautions
Bradycardia or heart block may occur in patients w/ & w/o known underlying cardiac conduction abnormalities. Monitor patients for symptoms of active or occult GI bleeding, especially patients at risk of developing ulcers eg, patients w/ history of ulcer disease or receiving concurrent NSAIDs. History of asthma or obstructive pulmonary disease. Pregnancy & lactation. Childn.
Adverse Reactions
Anorexia, diarrhea, fatigue, insomia, muscle cramps, nausea, vomiting. Abnormal dreams, arthritis, constipation, dizziness, ecchymosis, frequent urination, headache, mental depression, pain, drowsiness, fainting, wt loss.
Drug Interactions
Increased action w/ cholinomimetics/cholinesterase inhibitors eg, bethanechol succinylcholine, CYP450 3A4 & 2D6 inhibitors eg, ketoconazole, quinidine. Decreased action w/ CYP450 3A4 & 2D6 inhibitors eg, carbamazepine, dexamethasone, rifampicin, phenytoin.
