Omeprazole
Indications/Uses
Short-term treatment of duodenal ulcer, gastric ulcer, erosive reflux esophagitis/ulcerative. Zollinger-Ellison syndrome.
Dosage/Direction for Use
Duodenal ulcer 20 mg once daily for 4 wk. Severe cases: 40 mg once daily. Benign gastric ulcer 20 mg once daily for 8 wk. Severe cases: 40 mg once daily. Erosive reflux esophagitis 20 mg once daily for 4 wk, continued for further 4-8 wk if not fully healed. Reflux esophagitis refractory to other therapy 20 mg once daily. Zollinger-Ellison syndrome Initially 60 mg once daily. Severe disease & inadequate response to other therapy: 20-120 mg daily. Divide doses >80 mg & give bd.
Administration
Should be taken on an empty stomach: Take 30 min-1 hr before a meal.
Contraindications
Special Precautions
May be associated w/ increased risk of Clostridium difficle-associated diarrhoea, especially in hospitalised patients. Increased bone fracture risk in patients receiving high-, multiple daily doses & during long-term therapy (ie, ≥1 yr). Concomitant use w/ methotrexate; may consider temporary w/drawal of therapy during high-dose methotrexate administration. Exclude possibility of malignancy prior to treatment if gastric ulcer was suspected. Monitor Mg levels prior to treatment & periodically during prolonged treatment or concomitant intake w/ digoxin or hypomagnesaemia-causing drugs (eg, diuretics). Cutaneous lupus erythematosus (CLE) & SLE as both new onset & an exacerbation of existing autoimmune disease; avoid administration longer than medically indicated; discontinue use upon occurrence of signs or symptoms consistent w/ CLE or SLE. Pregnancy. Childn.
Adverse Reactions
Clostridium difficile-associated diarrhoea; fractures. Potentially, SLE & CLE.
Drug Interactions
May elevate & prolong serum levels of methorexate &/or its metabolite. Can delay elimination of diazepam, phenytoin & warfarin.
