Gliclazide
Dosage/Direction for Use
Initially 40-80 mg daily, adjust according to response up to 160 mg as a single dose. Max: 320 mg/daily.

Administration
Should be taken with food: Take w/ breakfast.
Contraindications
Hypersensitivity to gliclazide or other sulfonylureas or sulfonamides. IDDM particularly juvenile diabetes, diabetes complicated by ketosis & acidosis or diabetic pre-coma or coma. Severe hepatic or renal insufficiency. Concomitant treatment w/ miconazole, phenylbutazone, danazol & alcohol. Pregnancy & lactation. Childn.

Special Precautions
Hypoglycaemia, adrenal insufficiency, hypopituitarism, renal or hepatic insufficiency. Patients on β-blockers. Regular monitoring of blood & urine glucose. Elderly.

Adverse Reactions
Hypoglycaemia; GI disturbances, dermatological reactions; headache, nausea, dizziness, liver function abnormalities, cholestatic jaundice (discontinue drug), haematological effects including anemia, leucopenia & thrombocytopenia. Rarely, hepatitis.

Drug Interactions
Potentiated hypoglycaemic effect w/ aspirin, NSAIDs, phenylbutazone, clofibrate, sulfonamides, coumarin anticoagulants, cimetidine, imidazole antifungals, MAOIs. Diminished hypoglycaemic effect w/ rifampicin, barbiturates, phenytoin, alcohol, thiazide diuretics, dizoxide, glucocorticoids, estrogens or sympathomimetics. Early symptoms of hypoglycaemia may be masked by β-adrenoreceptor-blocking drugs eg, propranolol.
