Lacidipine
Indications/Uses
HTN either alone or in combination w/ other antihypertensive agents.
Dosage/Direction for Use
Initially 2 mg once daily taken at the same time each day; may be increased to 4 mg & if necessary to 6 mg.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Severe aortic stenosis.
Special Precautions
Preexisting abnormalities in SA & AV node activity, congenital or documented acquired QT prolongation, poor cardiac reserve, unstable angina pectoris, recent MI. Concomitant use w/ medications that prolong the QT interval eg, class I & III antiarrhythmics, TCAs, some antipsychotics, antibiotics (eg, erythromycin) & some antihistamines (eg, terfenadine). Hepatic impairment. Pregnancy & lactation. Childn.
Adverse Reactions
Headache, dizziness; palpitation, tachycardia; flushing; stomach discomfort, nausea; skin rash (erythema, itching); polyuria; asthenia, edema; reversible increase in alkaline phosphatase.
Drug Interactions
Additive hypotensive effect w/ antihypertensive agents (eg, diuretics, β-blockers, ACE inhibitors). May increase plasma levels w/ cimetidine. Altered bioavailability w/ grapefruit juice. Reverse decreased renal plasma flow & GFR induced by cyclosporin. Concurrent use w/ CYP3A4 inhibitors & inducers.
