Leuprorelin acetate
Indications/Uses
Management of endometriosis including pin relief & reduction of endometriotic lesions. In combination w/ norethindrone 5 mg daily for initial management of endometriosis & recurrence of symptoms. In concomitant w/ Fe therapy for the pre-op hematologic improvement of patients w/ anemia caused by uterine leiomyomata.
Dosage/Direction for Use
Endometriosis Duration of treatment: 6 mth. If symptoms recur after a course of therapy, consider retreatment w/ a 6-mth course. Uterine leiomyomata (fibroids) Duration of therapy: 3 mth.
Contraindications
Hypersensitivity to leuprorelin acetate, GnRH or GnRH agonists analogs. Undiagnosed abnormal vag bleeding. Pregnancy & lactation.
Special Precautions
Discontinue if sudden partial or complete loss of vision or if there is sudden onset of proptosis, diplopia or migraine. Thrombophlebitis & pulmonary embolism. Assessment & management of CV disease risk factors prior to initiation; women w/ risk factors including lipid abnormalities or cigarette smoking. May experience breakthrough bleeding in patients missing successive doses. May develop or worsen depression & occurrence of memory disorders. Discontinue if severe depression occurs. May cause loss in bone density over the course of treatment; risk factors eg, chronic alcohol &/or tobacco use, strong family history of osteoporosis or chronic use of drugs that can reduce bone mass (eg, anticonvulsants or corticosteroids).
Adverse Reactions
Asthenia, general pain, headache; hot flushes/sweats; nausea/vomiting, GI disturbances; edema, wt gain/loss; acne, hirsutism; joint disorder, myalgia; decreased libido, depression/emotional lability, dizziness, nervousness, neuromuscular disorders, paresthesias; skin reactions; breast changes/tenderness/pain; vaginitis.
Drug Interactions
May cause misleading results w/ diagnostic tests of pituitary gonadotropic & gonadal functions conducted during treatment & for up to 3 mth after discontinuation.
