Simvastatin
Indications/Uses
Prevention of CHD & treatment of hyperlipidemia.
Dosage/Direction for Use
CHD 10-20 mg/day as a single dose in the evening. Hyperlipidemia Start w/ 10 mg/day as a single daily dose in the evening. Dose adjustments should be made at intervals of not less than 4 wk to max of 80 mg/day.
Administration
May be taken with or without food: Avoid excessive consumption (>1 L/day) of grapefruit juice.
Contraindications
Active liver disease or unexplained persistent elevations of serum transaminases. Combination w/ mibefradil. Pregnancy & lactation. Childn.
Special Precautions
May cause myopathy especially in combination w/ gemfibrozil, fibrates. Liver function tests should be performed at start of therapy & periodically thereafter. Patients w/ history of liver disease. Alcohol.
Adverse Reactions
Abdominal pain, constipation & flatulence. Asthenia & headache, myopathy, nausea, diarrhea, rash, dyspepsia, pruritus, alopecia, dizziness, muscle cramps, myalgia, pancreatitis, paresthesia, peripheral neuropathy, vomiting & anemia.
Drug Interactions
Cyclosporins, itraconazole, ketoconazole, macrolides & nefazodone; fibric acid derivatives or niacin. Anticoagulants eg, warfarin.
