Colchicine
Indications/Uses
Relief of acute gout & for the prophylaxis of acute attacks particularly during the 1st few mth of treatment w/ allopurinol or uricosurics. Other conditions including amyloidosis, Behcet's syndrome, familial Mediterranean fever, ITP, pericarditis, primary biliary cirrhosis & pyoderma gangrenosum.
Dosage/Direction for Use
Acute gout 1 mg orally then 500 mcg every 2-3 hr.
Administration
Should be taken with food.
Special Precautions
Avoid in patients w/ blood disorders. Patients w/ cardiac, hepatic, renal or GI disease. Avoid during pregnancy. Elderly & debilitated patients.
Adverse Reactions
Diarrhoea, nausea, vomiting & abdominal pain. Profuse diarrhoea, GI haemorrhage, skin rashes, renal & hepatic damage in larger dose.
Drug Interactions
May cause colchicine toxicity w/ clarithromycin, erythromycin, or tolbutamide. Increased serum uric acid & interfere activity w/ thiazide diuretics. May cause muscle disorders w/ ciclosporin. May impair absorption of vit B12.
