Domperidone

Symptomatic relief of nausea & vomiting.

Adult & adolescent ≥12 yr & weighing ≥35 kg 1 tab up to tds. Max: 30 mg/day.

Should be taken on an empty stomach: Take 15-30 min before meals.

Hypersensitivity. Prolactinoma; conditions wherein stimulation of gastric motility could be harmful eg, GI haemorrhage, mechanical obstruction or perforation. Patients w/ known existing prolongation of cardiac conduction intervals, particularly QTc; significant electrolyte disturbances or underlying cardiac disease (eg, CHF). Co-administration w/ QT-prolonging drugs, potent CYP3A4 inhibitors (regardless of their QT-prolonging effects). Moderate or severe hepatic impairment.

Associated w/ CV effects. Promptly report any cardiac symptoms. Patients w/ lactose intolerance, galactosaemia or glucose/galactose malabsorption. May affect ability to drive & use machines. Patients w/ severe renal impairment. Pregnancy & lactation. Not recommended in childn <12 yr & adolescents weighing <35 kg. Elderly >60 yr.

Dry mouth. Loss of libido, anxiety, agitation, nervouness; dizziness, somnolence, headache, extrapyramidal disorder; diarrhea; rash, pruritus, urticaria; galactorrhoea, breast pain & tenderness; asthenia.

Should not be taken simultaneously w/ antacids or antisecretory drugs. Possible antagonised anti-dyspeptic effect w/ anticholinergic drugs. Contraindicated w/ QTc-prolonging medicinal drugs eg, anti-arrhythmics class IA (eg, disopyramide, hydroquinidine, quinidine), anti-arrhythmics class III (eg, amiodarone, dofetilide, dronedarone, ibutilide, sotalol), certain antipsychotics (eg, haloperidol, pimozide, sertindole), certain antidepressants (eg, citalopram, escitalopram), certain antibiotics (eg, erythromycin, levofloxacin, moxifloxacin, spiramycin), certain antifungal agents (eg, fluconazole, pentamidine), certain antimalarial agents (in particular halofantrine, lumefantrine), certain GI medicines (eg, cisapride, dolasetron, prucalopride), certain antihistaminics (eg, mequitazine, mizolastine), certain medicines used in cancer (eg, toremifene, vandetanib, vincamine), certain other medicines (eg, bepridil, diphemanil, methadone); apomorphine (unless benefit of co-administration outweighs the risks & recommended precautions are strictly fulfilled); potent CYP3A4 inhibitors eg, PIs (eg, ritonavir, saquinavir, telaprevir), systemic azole antifugals (eg, itraconazole, ketoconazole, posaconazole, voriconazole), certain macrolides (eg, clarithromycin, telithromycin). Not recommended w/ moderate CYP3A4 inhibitors (eg, diltiazem, verapamil & some macrolides). Caution w/ bradycardia & hypokalaemia-inducing drugs, azithromycin & roxithromycin.
