Indications/Uses
Advanced cancer of the ovary & small cell cancer of the lung.
Dosage/Direction for Use
Previously untreated adult w/ normal kidney function (CrCl >60 mL/min) 400 mg/m2 single short term IV dose administered by a 15-60 min infusion.
Contraindications
Hypersensitivity. Patients w/ fewer blood cells than normal; tumour that bleeds; severe kidney disease; planned or history of yellow fever vaccination.
Special Precautions
Caution in patients w/ symptoms of haemolytic uraemic syndrome; fever (≥38°C) or chills, which could be signs of infection; mild renal disease; latex sensitivity; elderly >65 yr; previous treatment w/ cisplatin or similar anti-cancer medicines. Risk of tumour lysis syndrome. Do not drive or use machines in case of any side effect that lessens such ability eg, nausea, vomiting, worsening of eyesight, or changes to vision & hearing. Possible risk of birth defects. Female patients of childbearing potential should take contraceptive measures before & during treatment w/ carboplatin. Men are advised not to father a child during, & up to 6 mth after treatment. May temporarily or permanently reduce fertility in men & women. Discontinue breastfeeding during treatment.
Adverse Reactions
Myelosuppression; tiredness, shortness of breath & paleness caused by anaemia; increased blood urea level; abnormal liver enzyme levels; nausea or vomiting; stomach pain & cramp; decrease in the blood level of Na, K, Ca, & Mg; decrease in renal CrCl. Diarrhoea or constipation; rash &/or itchy skin; ringing in the ears or changes in hearing; hair loss; flu-like symptoms; signs of infection; symptoms of severe allergic reaction; tingling or numbness in hands, feet, arms or legs; burning or prickling sensation; decreased tendon reflex; taste disturbance or loss of taste; temporary worsening of eyesight or changes to vision; heart disorders; tightness of the chest or wheezing; ILD; mucous membrane disorders; musculoskeletal disorder; problems w/ kidneys or urine; asthenia; increased blood level of bilirubin, creatinine, uric acid.
Drug Interactions
Concomitant use w/ medicines which can reduce the number of blood cells may require changes to the dosage & frequency of carboplatin treatment. Increased risk of kidney or hearing problems w/ aminoglycosides, vancomycin or capreomycin; some diuretics. Potential interaction w/ live or live-attenuated vaccines; immunosuppressants (eg, ciclosporin, tacrolimus, sirolimus). Concomitant use w/ blood thinning medicines eg, warfarin may require an increase in frequency of blood coagulation monitoring. Increased risk of seizure w/ phenytoin & fosphenytoin. Carboplatin may affect liver's ability to cope w/ alcohol.
