Rescue therapy after treatment w/ ave to high dose of MTX. In the treatment of megaloblastic anemia owing to folate deficiency. In combination w/ 5-FU for the treatment of metastasised colorectal carcinoma.
Dosage/Direction for Use
Rescue therapy Begin 24 hr after termination of MTX therapy. Usually 15 mg 3 hrly IV for 1st 24 hr. Then, one 15 mg tab 6 hrly for 48 hr. Megaloblastic anemia Initially 10 mg daily for 10-15 days, then adjust dosage according to hematological response. Metastasised colorectal carcinoma Wkly schedule: 20-200 mg/m2 in 2 hr infusion w/ 500 mg/m2 5-FU as bolus inj. Mthly schedule: 20 or 200 mg/m2 bolus immediately followed by 425 or 370 mg/m2 5-FU bolus once a mth for 5 consecutive days.
Administration
May be taken with or without food.
Contraindications
Undiagnosed megaloblastic anemia w/ inadequate vit B12.
