Anastrozole
Indications/Uses
Adjuvant treatment of postmenopausal women w/ hormone receptor +ve early invasive breast cancer. Advanced breast cancer in postmenopausal women w/ hormone receptor +ve or hormone receptor unknown locally advanced or metastatic breast cancer.
Dosage/Direction for Use
Adult including elderly 1 mg once daily. Recommended treatment duration for early disease: 5 yr.
Administration
May be taken with or without food: Swallow whole, do not chew/crush.
Contraindications
Hypersensitivity. Premenopausal women. Co-administration w/ oestrogen-containing therapies. Concurrent tamoxifen therapy. Severe renal impairment (CrCl <20 mL/min). Moderate or severe hepatic disease. Pregnancy & lactation.
Special Precautions
Biochemically define menopause in any patient where there is doubt about menopausal status. Formally assess bone mineral density by bone densitometry (eg, DEXA scanning) in women w/ or at risk of osteoporosis at the commencement of treatment & at regular intervals thereafter; carefully monitor & initiate appropriate treatment or prophylaxis for osteoporosis. Not to be used outside clinical trials w/ LHRH analogues. May reduce bone mineral density w/ a possible consequent increased risk of fracture; consider use of bisphosphonates in postmenopausal women. Possible asthenia & somnolence; observe caution when driving or operating machinery. Not recommended for use in childn.
Adverse Reactions
Hot flushes; asthenia; arthralgia/joint stiffness, arthritis, osteoporosis; headache; nausea; rash; depression. Vag dryness & bleeding; alopecia, allergic reactions; diarrhoea, vomiting; somnolence, carpal tunnel syndrome, sensory disturbances; increased alkaline phosphatase, ALT & AST; anorexia, hypercholesterolaemia; bone pain, myalgia.
Drug Interactions
Negation of pharmacological action w/ oestrogen-containing therapies. Pharmacological action may be diminished by tamoxifen.
