Amlodipine
Indications/Uses
1st-line mono- or combination therapy for HTN. Mono- or combination therapy for symptomatic treatment of chronic stable angina; confirmed or suspected vasospastic angina (Prinzmetal's or variant angina).
Dosage/Direction for Use
HTN & angina Initially 5 mg once daily, may be increased to max: 10 mg. CAD 5-10 mg once daily.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity to amlodipine or other dihydropyridines. Cardiogenic shock, clinically significant aortic stenosis, unstable angina (excluding Prinzmetal's angina). Pregnancy & lactation.
Special Precautions
Pulmonary oedema in patients w/ NYHA III & IV heart failure of nonischaemic aetiology. Hepatic impairment. Childn.
Adverse Reactions
Flushing; dizziness, headaches, somnolence; abdominal pain, nausea; palpitations; fatigue, oedema.
Drug Interactions
Affects trough conc of cyclosporin. Increased tacrolimus blood levels. Increased exposure to simvastatin. Increased systemic exposure w/ diltiazem. Increased plasma conc by strong CYP3A4 inhibitors eg, ketoconazole, itraconazole, ritonavir. Increased risk of hypotension w/ clarithromycin. Decreased plasma conc by CYP3A4 inducers eg, rifampicin, Hypericum perforatum.
