Indications/Uses
Reduces of LDL-cholesterol, apolipoprotein B & triglycerides & increase HDL-cholesterol in the management of hyperlipidaemias, including primary hypercholesterolaemia (type IIa), mixed dyslipidaemia (type IIb), & hypertriglyceridaemia (type IV), as well as in patients w/ homozygous familial hypercholesterolaemia. Reduces the progression of atherosclerosis.
Dosage/Direction for Use
Initially 5-10 mg once daily at night. Maintenance dose: 5-40 mg once daily. Patient w/ high CV risk 40 mg dose.
Administration
May be taken with or without food.
Contraindications
Active liver disease including unexplained persistent elevations of hepatic aminotransaminases. Pregnancy.
Special Precautions
Discontinue use if marked or persistent increases in serum aminotransferase or creatine phosphokinase conc occurs.
Adverse Reactions
GI disturbances; headache, skin rashes, dizziness, blurred vision, insomnia, dysgeusia; reversible increase in serum aminotransferase conc; myopathy.
Drug Interactions
Increased risk of myopathy w/ fibrates, gemfibrozil & nicotinic acid. Limited metabolism w/ enzyme inhibitors eg, simvastatin. Increased plasma conc w/ ciclosporin, HIV PIs & to a lesser extent w/ gemfibrozil.
