Indications/Uses
Management of peripheral vascular disease.
Dosage/Direction for Use
Administration
Should be taken on an empty stomach: Take at least 30 min before or 2 hr after breakfast or dinner.
Contraindications
Patients w/ heart failure of any severity & CHF. Patients receiving ≥2 additional antiplatelet/anticoagulant agents.
Special Precautions
Patients w/ history of ischemic heart disease. Monitor patients for the development of a new systolic murmur or cardiac symptoms after initiating therapy. Periodically monitor platelet & WBC counts. Avoid use in patients w/ hemostatic disorders or active pathological bleeding. Discontinue use in cases of retinal bleeding. Higher risk of intraocular bleeding in patients w/ diabetes. Perform full blood count if infection is suspected or there is any other clinical evidence of blood dyscrasia. Immediately discontinue use if there is clinical or laboratory evidence of hematologic abnormalities. Surgery including minor invasive procedures eg, tooth extraction; discontinue therapy 5 days before surgery. May affect ability to drive & use machines. Moderate or severe hepatic impairment. Severe renal impairment (CrCl <25 mL/min). Pregnancy & lactation. Childn.
Adverse Reactions
Drug Interactions
Increased inhibition of ADP-induced ex vivo platelet aggregation w/ aspirin. Clopidogrel & other antiplatelet agents; warfarin & other anticoagulant agents. Increased Cmax & AUC w/ strong (eg, ketoconazole, itraconazole, fluconazole, miconazole, fluvoxamine, fluoxetine, nefazodone, sertraline) & moderate (eg, erythromycin & other macrolides, diltiazem, grapefruit juice) CYP3A4 inhibitors. Decreased Css, Cmax & AUC w/ lovastatin. May alter antiplatelet effect w/ CYP3A4 & CYP2C19 inducers (eg, carbamazepine, phenytoin, rifampicin, St. John's wort).
