Lymecycline
Indications/Uses
Infections due to microorganisms sensitive to tetracyclines; acne.
Dosage/Direction for Use
Acne 300 mg daily; usual treatment duration is 12 wk. Indications other than acne Adult 600 mg daily.
Administration
Should be taken on an empty stomach: Take 1 hr before or 2 hr after meals.
Contraindications
Hypersensitivity to tetracyclines. Concurrent treatment w/ oral retinoids. Childn <8 yr due to risk of permanent dental staining & enamel hypoplasia.
Special Precautions
May cause oesophageal irritation & ulceration. Impaired renal or hepatic functions. Overdosage could result in hepatotoxicity; high dosage may be nephrotoxic. Avoid exposure to direct sunlight & UV light. Discontinue use if erythematous cutaneous manifestations occur; any evidence of raised ICP develop during treatment. May lead to renal tubular acidosis (Pseudo-Fanconi syndrome) in expired tetracycline use. Prolonged use may result in appearance of resistant organisms & superinfection. Exacerbation of SLE. Caution in treatment of patients w/ myasthenia gravis who may be at risk of worsening condition. False +ve urine glucose determinations; interfere w/ fluorometric determinations of urine catecholamines resulting in falsely increased values (Hingerty's method). Not to be administered during pregnancy & lactation.
Adverse Reactions
Nausea, abdominal pain, diarrhea; headache. Dental dyschromia &/or enamel hypoplasia especially in childn <8 yr. Haemolytic anaemia, eosinophilia & other hematologic disorders. Extra-renal hyperazotemia linked to an anti-anabolic effect which may be intensified by association w/ diuretics. SLE. Pancreatitis.
Drug Interactions
Risk of intracranial HTN w/ oral retinoids & vit A (>10,000 IU/day). May decrease absorption w/ Fe prep & antacids, Mg/Al & Ca hydroxides, oxides & salts, activated charcoal, cholestyramine, bismuth chelates & sucralfate. May reduce absorption of tetracycline w/ medicinal products which increase gastric pH. Enzyme inducers eg, barbiturates, carbamazepine, phenytoin may accelerate decomposition. Effect of oral coumarin-type anticoagulants could be increased w/ increased risk of haemorrhage. May interfere w/ bacteriocidal action of penicillin & β-lactam antibiotics. Fatal renal toxicity w/ methoxyflurane. Decreased digestive absorption due to increased gastric pH w/ didanosine. Increase in serum lithium levels.
