Bicalutamide
Indications/Uses
Prostatic cancer; in combination w/ gonadorelin analogue for palliative treatment of advanced prostatic cancer.
Dosage/Direction for Use
150 mg daily. May be given as monotherapy or adjuvant therapy to surgery or radiotherapy in men w/ locally advanced disease.
Administration
Should be taken with food.
Contraindications
Severe hepatic impairment.
Special Precautions
Patients w/ CV disease due to possibility of fluid retention. Regular liver function testing. Hepatic impairment. Stop therapy or reduced dosage if there is evidence of hepatotoxicity.
Adverse Reactions
Hot flushes & reversible gynaecomastia or breast tenderness, galactorrhoea. Nausea, vomiting, diarrhoea, increased appetite, anorexia, & sleep disturbances. Epidermal necrolysis, & liver damage. Anaemias, haemolysis, headache, dizziness, malaise, blurred vision, anxiety, depression, decreased libido, impotence, & HTN. Discoloration of the urine to amber or yellow-green. Pruritus, asthenia, alopecia, hair regrowth, & dry skin.
Drug Interactions
Terfenadine, astemizole, & cisapride & other drugs w/ narrow therapeutic index that are metabolised by cytochrome P450 isoenzymes. May displace warfarin from protein binding sites.
