Indications/Uses
Peripheral & central neuropathic pain in adults. Adjunctive therapy in adults w/ partial seizures w/ or w/o secondary generalisation. Generalised anxiety disorder in adults.
Dosage/Direction for Use
150-600 mg daily in 2 or 3 divided doses. Neuropathic pain Initially 150 mg daily in 2 or 3 divided doses. May be increased to 300 mg daily after an interval of 3-7 days if needed. Max: 600 mg daily after an additional 7-day interval. Epilepsy Initially 150 mg daily in 2 or 3 divided doses. May be increased to 300 mg daily after 1 wk. Max: 600 mg daily after an additional wk. Generalised anxiety disorder 150-600 mg daily in 2 or 3 divided doses. Initially 150 mg daily. May be increased to 300 mg daily after 1 wk. Following an additional wk dose may be increased to 450 mg daily. Max: 600 mg daily after an additional wk. Discontinuation of pregabalin Gradually over min of 1 wk independent of indication. Renal impairment CrCl ≥60 mL/min Initially 150 mg daily. Max: 600 mg daily in 2 or 3 divided doses. CrCl ≥30-<60 mL/min Initially 75 mg daily. Max: 300 mg daily in 2 or 3 divided doses. CrCl ≥15-<30 mL/min Initially 25-50 mg daily. Max: 150 mg daily in 2 divided doses or once daily. CrCl <15 mL/min Initially 25 mg daily. Max: 75 mg once daily. Supplementary dose following hemodialysis Initially 25 mg daily. Max: 100 mg given immediately after each 4-hr session.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Hypersensitivity reactions. Diabetic patients; CHF. Dizziness, somnolence, loss of consciousness, confusion & mental impairment. Vision-related effects including loss of vision, visual blurring or other changes of visual acuity. W/drawal of concomitant antiepileptic medicinal products. W/drawal symptoms eg, insomnia, headache, nausea, anxiety, diarrhea, flu syndrome, nervousness, depression, pain, convulsion, hyperhidrosis & dizziness; convulsions including status epilepticus & grand mal convulsions during use or shortly after discontinuation. Increased CNS adverse reactions & somnolence in treatment of central neuropathic pain due to spinal cord injury. Monitor signs of suicidal ideation & behaviors. Reduced lower GI function; concomitant use w/ opioid analgesics. History of substance abuse & monitor symptoms of misuse, abuse or dependence. May precipitate encephalopathy in patients w/ underlying conditions. Contains lactose monohydrate; galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption. May influence ability to drive or use machines. Renal failure. Pregnancy & lactation. Childn <12 yr & adolescent 12-17 yr. Elderly.
Adverse Reactions
Dizziness, somnolence, headache. Nasopharyngitis; increased appetite; euphoric mood, confusion, irritability, disorientation, insomnia, decreased libido; ataxia, abnormal coordination, tremor, dysarthria, amnesia, memory impairment, disturbance in attention, paraesthesia, hypoaesthesia, sedation, balance disorder, lethargy; blurred vision, diplopia; vertigo; vomiting, nausea, constipation, diarrhea, flatulence, abdominal distension, dry mouth; muscle cramp, arthralgia, back pain, pain in limb, cervical spasm; erectile dysfunction; edema, peripheral edema, abnormal gait, fall, feeling drunk, feeling abnormal, fatigue; increased wt.
Drug Interactions
May potentiate the effects of ethanol & lorazepam. Resp failure & coma w/ CNS depressant medicinal products. Additive in the impairment of cognitive & gross motor function w/ oxycodone.
