Indications/Uses
Tab/Susp: Trichomoniasis, amoebiasis, giardiasis, Vincent's infection. Prevention & treatment of anaerobic infections. Inj Treatment of anaerobic bacterial infections & protozoal infections eg, trichomonal vaginalis, bacterial vaginosis & Entamoeba histolytica & Giardia lamblia infections. Surgical & gynecological sepsis. Treatment of antibiotic-associated colitis & established cases of tetanus.
Dosage/Direction for Use
Tab Adult Amoebic dysentery Initially 2 g as single dose for 2 days then 500 mg tid for 5-10 days. Hepatic & other extraintestinal amoebiasis 500-800 mg tid or 2 g as single dose for 2 days. Giardiasis 2 g daily as single dose for 3 successive days. Childn 7-10 yr ½ adult dose. Susp Childn ≥10 yr 1-2 tsp, 5-10 yr 2 tsp, 2-5 yr 1 tsp. To be taken 8 hrly. Inj Adult & childn >12 yr 100 mL IV infusion 8 hrly for 7 days immediately before, during or after an operation by the same dose 8 hrly until oral medication can be given to complete a 7-day course. Treatment Adult & childn >12 yr 100 mL IV infusion 8 hrly for 7 days. Childn <12 yr As for adults, but the single IV dose is based on 1.5 mL/kg.
Administration
Tab: Should be taken with food. Susp: Should be taken on an empty stomach: Take at least 1 hr before meals.
Contraindications
Inj: Patients w/ blood dyscrasias or w/ active CNS disease. Pregnancy.
Special Precautions
Blood dyscrasias, active CNS disease.
Adverse Reactions
Tab/Susp: GI discomfort, anorexia, nausea, coated tongue, dry mouth & unpleasant taste, headache, pruritus & skin rash; vomiting, diarrhea, weakness, vertigo, ataxia, depression & darkening of the urine. Occasionally, temporary moderate leukopenia. Peripheral neuropathy on prolonged treatment. Inj: Convulsive seizures & peripheral neuropathy. Nausea, headache, anorexia & occasionally vomiting, diarrhea, epigastric distress & abdominal cramping. Constipation.
Drug Interactions
Disulfiram-like effect w/ alcohol. Enhances anticoagulant effect of warfarin.
