Contents
Pantoprazole Na sesquihydrate 40 mg, domperidone 30 mg
Indications/Uses
Symptomatic management of burning in various acid peptic disorders associated w/ nausea & vomiting; GERD, non-ulcer dyspepsia, gastroparesis & idiopathic hiccup or any other peptic disease accompanied by nausea &/or vomiting.
Dosage/Direction for Use
Administration
Should be taken on an empty stomach: Swallow whole, do not chew.
Contraindications
Hypersensitivity. Pantoprazole: Hepatic impairment. Domperidone: History of hypersensitivity to dimenhydrinate or related compd. GI haemorrhage, obstruction & perforation, or after surgery. Pregnancy.
Special Precautions
Renal or hepatic dysfunction. Pregnancy & lactation. Not recommended in child <12 yrs. Long-term therapy may lead to bacterial overgrowth in the GIT. Domperidone: Increases serum prolactin levels resulting to galactorrhoea in females & gynaecomastia in males. Hypertensive crisis in patients w/ pheochromocytoma.
Adverse Reactions
Rash; headache; diarrhoea; pruritus; dizziness. Pantoprazole: GIT infections; anaphylaxis, angioedema, chest pain, dyspnoea, erythema multiforme, gastroenteritis, hyperglycaemia, infection, inj site reaction, jaundice, optic neuropathy, anterior ischaemia, pancreatitis, speech disorder. Domperidone: Serum prolactin level may rise resulting in galactorrhea in females & less frequently gynaecomastia in males due. Dry mouth, thirst, insomnia, lethargy, irritability, GI disturbances, hot flushes, mastalgia, menstrual irregularities, urticaria, stomatitis, conjunctivitis, urinary frequency, dysuria, oedema, palpitations, leg cramps, asthenia, drug intolerance; drowsiness, & itching.
Drug Interactions
Pantoprazole: May interfere w/ the absorption of drugs where gastric pH is an important determinant of bioavailability (eg, ketoconazole, ampicillin esters, & Fe salts). Domperidone: May inhibit the anti-dyspeptic effects w/ anti-cholinergic drugs. Antagonized effect w/ antimuscarinic agents & opioid analgesics. Suppressed peripheral effects (eg, digestive disorders, nausea & vomiting) of dopaminergic agonists. May influence the absorption w/ orally administered drugs particularly those w/ SR or EC formulations. Interferes w/ serum prolactin levels & w/ other hyperprolactinaemic agents w/ some diagnostic tests. Lowered bioavailability w/ antacids & anti-secretory agents. Decreased oral bioavailability w/ antacids & anti-secretory agents.
