Indications/Uses
Infections due to Staph resistant to benzylpenicillin. Bone & joint infections, endocarditis, peritonitis (associated w/ continues ambulatory peritoneal dialysis), pneumonia, skin disorders (including soft-tissue infections, surgical infection prophylaxis, & toxic shock syndrome).
Dosage/Direction for Use
500 mg-1 g IM or slow IV infusion every 4-6 hr. Childn <40 kg 50-100 mg/kg daily in divided doses.
Contraindications
Special Precautions
Patients w/ history of allergy, especially to drugs. Risk of neurotoxicity. Avoid intrathecal route; skin contact w/ penicillins. Monitor renal, hepatic & haematological status during prolonged & high-dose therapy. Patients w/ spirochaete infections, particularly syphilis. Pregnancy.
Adverse Reactions
Hypersensitivity reactions eg, skin rashes. Haemolytic anaemia & neutropenia; prolongation of bleeding time & defective platelet function; convulsions & other signs of CNS toxicity. Jarisch-Herxheimer reaction in patients w/ syphilis. GI effects like diarrhea & nausea.
Drug Interactions
Prolonged t½ w/ probenecid. May interact w/ chloramphenicol & tetracyclines.
