Indications/Uses
Duodenal & gastric ulcer. Treatment/prophylaxis of reflux oesophagitis. Eradication of H. pylori concurrently given w/ appropriate antibiotic therapy for the treatment of H pylori ulcers. NSAID-associated benign gastric & duodenal ulcers in patients requiring continued NSAID treatment. Prophylaxis of NSAID-associated gastric ulcers & duodenal ulcers. GERD. Zollinger-Ellison syndrome.
Dosage/Direction for Use
Duodenal ulcer 30 mg once daily for 2 wk. Gastric ulcer, reflux oesophagitis, NSAID associated benign gastric & duodenal ulcers in patients requiring continued NSAID treatment 30 mg once daily for 4 wk. Prophylaxis of reflux oesophagitis 15 mg once daily, may be increased up to 30 mg daily. Eradication of H. pylori 30 mg lansoprazole bid for 7 days + clarithromycin 250-500 mg bid + amoxicillin 1 g daily or 30 mg lansoprazole + clarithromycin 250 mg bid + metronidazole 400-500 mg bid. NSAID associated gastric & duodenal ulcers in patients at risk (>65 yr or history of gastric or duodenal ulcer) requiring prolonged NSAID treatment 15 mg once daily, may increase to 30 mg once daily. Symptomatic GERD 15 or 30 mg daily. Zollinger-Ellison syndrome Initially 60 mg once daily. Dose should be individually adjusted & continue treatment as long as necessary. Elderly Max: 30 mg once daily.
Administration
Should be taken on an empty stomach.
Contraindications
Hypersensitivity. Do not administer w/ atazanavir.
Special Precautions
Exclude possibility of malignant gastric tumour. Patients w/ moderate & severe hepatic dysfunction. Severe hypomagnesaemia. Subacute cutaneous lupus erythematosus. Increased chromagranin A (CgA) level. Fructose intolerance, glucose-galactose malabsorption or sucrase-isomaltase insufficiency. Pregnancy & lactation.
Adverse Reactions
Headache, dizziness; nausea, diarrhoea, stomachache, constipation, vomiting, flatulence, dry mouth or throat, benign fundic gland polyps. Increased liver enzyme levels. Urticaria, itching, rash. Fatigue.
Drug Interactions
Atazanavir. Ketoconazole & itraconazole. Increased digoxin plasma levels. Reduced plasma conc of theophylline. Increased plasma conc of tacrolimus, fluvoxamine. Decrease bioavailability w/ sucralfate/antacids.
