Indications/Uses
Peripheral & central neuropathic pain in adults. Adjunctive therapy in adults w/ partial seizures w/ or w/o secondary generalisation. Generalised anxiety disorder (GAD) in adults.
Dosage/Direction for Use
150-600 mg/day given in 2 or 3 divided doses. Neuropathic pain Initially 150 mg/day in 2 or 3 divided doses, may be increased to 300 mg/day after an interval of 3-7 days. Max: 600 mg/day after an additional 7-day interval. Epilepsy Initially 150 mg/day in 2 or 3 divided doses, may be increased to 300 mg/day after 1 wk. Max: 600 mg/day after an additional wk. GAD Initially 150 mg/day, may be increased to 300 mg/day after 1 wk. Following an additional wk, dose may be increased to 450 mg/day. Max: 600 mg/day after an additional wk. Renal impairment CrCl ≥60 mL/min Initially 150 mg/day bid or tid, max: 600 mg/day, ≥30-<60 mL/min Initially 75 mg/day bid or tid, max: 300 mg/day; ≥15-<30 mL/min Initially 25-50 mg/day once daily or bid, max: 150 mg/day, <15 mL/min Initially 25 mg/day once daily, max: 75 mg/day. Supplementary dosage following haemodialysis: Initially 25 mg as a single dose, max: 100 mg/day.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Diabetic patients. Discontinue immediately if symptoms of angioedema eg, facial, perioral or upper airway swelling occur. Dizziness, somnolence, loss of consciousness, confusion & mental impairment. Vision-related effects. Renal failure. W/drawal symptoms. CHF in elderly CV compromised patients. Increased incidence of AR in treatment of central neuropathic pain due to spinal cord injury & additive effect w/ concomitant medicinal products (eg, anti-spasticity agents). Suicidal ideation & behaviour. Reduced lower GIT function. History of substance abuse & monitor for symptoms of pregabalin misuse, abuse or dependence. May precipitate encephalopathy. May have minor or moderate influence on the ability to drive & use machines. Women of childbearing potential/contraception in males & females. Pregnancy & lactation.
Adverse Reactions
Dizziness, somnolence, headache. Nasopharyngitis; increased appetite; euphoric mood, confusion, irritability, disorientation, insomnia, decreased libido; ataxia, abnormal coordination, tremor, dysarthria, amnesia, memory impairment, disturbance in attention, paraesthesia, hypoaesthesia, sedation, balance disorder, lethargy; blurred vision, diplopia; vertigo; vomiting, nausea, constipation, diarrhoea, flatulence, abdominal distension, dry mouth; muscle cramp, arthralgia, back pain, pain in limb, cervical spasm; erectile dysfunction; peripheral oedema, oedema, abnormal gait, fall, drunk feeling, abnormal feeling, fatigue; increased wt.
Drug Interactions
May potentiate the effects of ethanol & lorazepam.
