Pregabalin
Indications/Uses
Generalized anxiety disorder in adult. Adjunctive therapy for partial onset seizures.
Dosage/Direction for Use
150-600 mg daily in 2 or 3 divided doses. Epilepsy Initially 150 mg daily as 2 or 3 divided doses, may be increased to 300 mg daily after 1 wk. Max: 600 mg daily after an additional wk. Generalized anxiety disorder Initially 150 mg daily, may be increased to 300 mg daily after 1 wk; may be increased to 450 mg daily after an additional wk. Max: 600 mg daily after an additional wk.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Increased risk of suicidal thoughts or behavior; worsening signs & symptoms of depression, unusual mood changes or thoughts about self-harm. Discontinue use if angioedema, hypersensitivity; myopathy is diagnosed or suspected; markedly elevated creatine kinase levels. Increased risk of accidental injury (fall) in elderly. Patients w/ history of substance abuse, underlying conditions that may precipitate encephalopathy; previous episode of angioedema & those taking other drugs associated w/ angioedema; CHF. May cause peripheral edema & wt gain; convulsions (during or shortly after treatment). Frequent assessment for those who already routinely monitored for ocular conditions & if visual disturbance persists. May affect ability to drive or operate machinery. Pregnancy & lactation. Childn <18 yr.
Adverse Reactions
Asthenia, body odor, chills, death, sudden death, edema (generalized, face, peripheral), fatigue, abnormal feeling, drunk, fever, hangover effect, abnormal healing, hernia, hypothermia, joint swelling, malaise, mucous membrane disorder, pain (pelvic, neck, back, chest, limb), pain in extremities & decreased tolerance.
Drug Interactions
Potentiate effects of ethanol & lorazepam. Additive in cognitive & gross motor impairment caused by oxycodone. Reduced lower GI function w/ opioid analgesics eg, oxycodone, morphine or codeine. Increased risk of developing angioedema w/ ACE inhibitors.
