Indications/Uses
RA & OA. Adjunctive treatment of familial adenomatous polyposis (FAP).
Dosage/Direction for Use
OA 200 mg/day as single or 2 divided doses. RA 100-200 mg bid. Pain & dysmenorrhea Initially 400 mg followed by additional dose of 200 mg if necessary on the 1st day then 200 mg bid thereafter. FAP 400 mg bid.
Administration
May be taken with or without food: Dose for OA/RA may be given w/ or w/o meals, but doses for familial adenomatous polyposis must be given w/ meals.
Contraindications
Severe heart failure, inflammatory bowel disease, asthma, urticaria. Patients who have a history of stroke, CVA, heart attack, MI, CABG, uncontrolled HTN, CHF, NYHA II-IV. Renal impairment (CrCl <30 mL/min).
Special Precautions
Hypersensitivity to sulfonamides. W/draw therapy if hepatic toxicity develops. Dehydration, anemia, preexisting asthma. Liver dysfunction, impaired renal function. Pregnancy.
Adverse Reactions
Constipation, diverticulitis, dysphagia, eructation, esophagitis, gastritis, gastroesophageal reflux, hemorrhoids, hiatal hernia, melena, dry mouth, stomatitis, tenesmus, tooth disorder, vomiting; aggravated HTN, angina pectoris, CAD, MI; increased hepatic function & SGOT/SGPT; anorexia, anxiety, increased appetite, depression, nervousness, somnolence; albuminuria, cystitis, dysuria, hematuria, micturition frequency, renal calculus, urinary incontinence, UTI.
Drug Interactions
Changes in plasma conc w/ CYP2C9 inhibitors. Increased plasma conc w/ fluconazole. CYP2D6 inhibitors.
