Indications/Uses
Infections caused by penicillinase-producing staphylococci; resp tract infections; skin & soft-tissue infections; disseminated infections caused by pneumococci, group A β-hemolytic streptococci & penicillin G-resistant & -sensitive staphylococci.
Dosage/Direction for Use
Adult 250 mg tid or 500 mg bid. Childn 125 mg tid. Resp tract infection 3 g bid. Acute UTI 3 g repeated once 12 hr later.
Administration
Should be taken on an empty stomach.
Contraindications
Hypersensitivity to penicillins or cephalosporins. Not to be given to patients w/ a history of penicillin allergy. Incompatible w/ aminoglycosides, tetracyclines, erythromycin & polymyxin B. Not to be administered to neonates born of mothers who are hypersensitive to penicillin.
Special Precautions
Generalized sensitivity reaction can develop w/in few hr or wk of commencing treatment. Supra-infection w/ C. albicans, other fungi or organisms may occur. Risk of neurotoxicity & CHF in administration of high doses in patients w/ renal impairment. Electrolyte balance disturbance may occur following administration of large doses. Increased liver enzymes. Monitor renal & haematological systems during prolonged & high-dose therapy. Patients w/ syphilis may exhibit Jarisch-Herxheimer reaction. Use a skin test for sensitivity to determine patients who are most likely to develop allergic reactions to penicillin.
Adverse Reactions
Hypersensitivity reaction eg, diarrhoea, nausea & vomiting.
Drug Interactions
Inhibited serum binding w/ aspirin, sulphamethoxypyridazine & sulphaethidole.
