Amlodipine besilate
Indications/Uses
Management of HTN & prophylaxis of angina.
Dosage/Direction for Use
HTN & angina Initially 5 mg once daily. Max dose: 10 mg once daily.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity to amlodipine & other dihydropyridines.
Special Precautions
Hypotension, poor cardiac reserve, heart failure, hepatic impairment. Pregnancy & lactation.
Adverse Reactions
Abdominal pain, nausea; palpitation, flushing, edema; headache, dizziness, sleep disturbances, fatigue.
Drug Interactions
Metabolism inhibited w/ macrolides, azole antifungals & HIV-PIs. Reduced effects w/ carbamazepine, phenobarb & phenytoin. Decrease renal & nonrenal clearance of digoxin. Increased bioavailability w/ grapefruit juice. Gingival hyperplasia exacerbated w/ ciclosporin. Excessive myocardium depression w/ β-adrenergic blockers. Dangerous hypotensive effects w/ prazosin, methyldopa & clonidine. Increased action w/ nondepolarizing muscle relaxants. Decreased 1st-pass effects w/ cimetidine.
