Indications/Uses
Advanced ovarian carcinoma of epithelial origin in 1st- & 2nd-line therapy, after other treatments have failed. Small cell carcinoma of the lung.
Dosage/Direction for Use
Previously untreated adult patient w/ normal renal function 400 mg/m2 as short-term IV infusion (15-60 min). Therapy should not be repeated until 4 wk after previous carboplatin course.
Contraindications
Severe preexisting renal impairment, myelosuppression. Severe allergic reactions to carboplatin or other platinum-containing compd.
Special Precautions
Monitor hematologic & renal function tests. Perform blood counts prior to therapy & at wkly intervals thereafter. Neurological evaluation & assessment of hearing should also be performed on a regular basis. Long-term therapy. Abnormal kidney function or concomitant therapy w/ drugs w/ nephrotoxic potential. Frequent monitoring of peripheral blood counts throughout & following therapy. Manifestation or exacerbation of infections & bleeding tendency. Women of childbearing potential. Childn. Elderly.
Adverse Reactions
Bone marrow suppression, thrombocytopenia, leucopenia, myelosuppression, infections, hemorrhagic complications; increased blood urea & serum creatinine; nausea, vomiting; rash, urticaria, erythema, pruritus; hearing loss; peripheral neuropathies; abnormal liver function test; pain, asthenia, alopecia, cardiac failure, embolism, cerebrovascular accidents.
Drug Interactions
Increased nephrotoxicity w/ aminoglycoside antibiotics. Concomitant use w/ other antineoplastic agents or radiotherapy.
