Mercaptopurine
Indications/Uses
Treatment or maintenance therapy in acute leukemia, acute lymphoblastic leukemia & acute myelogenous leukemia. Chronic granulocytic leukemia.
Dosage/Direction for Use
Adult & childn 2.5 mg/kg/day. Maintenance: 1.5-2.5 mg/kg/day as single dose.
Administration
Should be taken on an empty stomach: Best taken 1 hr before or 2 hr after meals. Ensure adequate fluid intake. Avoid taking w/ milk-rich products.
Special Precautions
Full blood counts must be taken daily during remission induction. Monitor liver function tests & uric acid levels in blood & urine regularly. Interrupt treatment at the 1st sign of an abnormal large fall in leukocyte & platelet count. Discontinue if jaundice becomes apparent. Pregnancy & lactation.
Adverse Reactions
Bone marrow suppression. Hepatotoxicity. Hepatic necrosis & biliary stasis. Anorexia, nausea, vomiting & oral ulceration occasionally occur.
Drug Interactions
Decreased rate of catabolism w/ allopurinol. Inhibition of anticoagulant effect of warfarin.
