Reduce LDL-cholesterol, apolipoprotein B & increase HDL-cholesterol in the treatment of hyperlipidemias including hypercholesterolemias & combined hyperlipidemia, hypertriglyceridemia & dysbetalipoproteinemia. Adjunctive therapy in patients w/ homozygous familial hypercholesterolemia who have some LDL-receptor function.
Usual dose: 10-20 mg once daily. Patient requiring large reduction in LDL-cholesterol Initially 40 mg daily. Dose may be adjusted at 4-wk intervals up to a max of 80 mg daily. Childn & adolescent 10-17 yr w/ heterozygous familial hypercholesterolemia Initially 10 mg once daily, adjusted to a max of 20 mg once daily.
May be taken with or without food: Avoid excessive consumption (>1 L/day) of grapefruit juice.
Contraindications
Acute liver disease or unexplained persistently raised serum aminotransferase conc.
Special Precautions
Assess liver functions before treatment & monitor periodically 1 yr after the last dose elevation. Patients w/ severe renal impairment. Risk of reversible increases in serum aminotransferase conc. Pregnancy.
