Indications/Uses
Luteal support as part of an assisted reproductive technology treatment program for infertile women.
Dosage/Direction for Use
Adult 100 mg tds starting at oocyte retrieval, continued for up to 10 wk total duration or 12 wk of gestation.
Contraindications
Hypersensitivity. Known or suspected breast or genital tract cancer; active or history of arterial or VTE or severe thrombophlebitis; porphyria; undiagnosed vag bleeding. Severe hepatic dysfunction or disease. Known missed abortion or ectopic pregnancy.
Special Precautions
Discontinue use if MI, cerebrovascular disorders, arterial or VTE (VTE or pulmonary embolism), thrombophlebitis or retinal thrombosis are suspected; transient ischemic events, appearance of sudden severe headaches or papillary edema- or retinal hemorrhage-related vision impairments occur. History of depression. Epilepsy, migraine, asthma, cardiac dysfunction. Diabetic patients; patients >35 yr, smokers, w/ risk factors for atherosclerosis. Assess infertility causes prior to treatment. Avoid abrupt discontinuation. May affect ability to drive & use machines. Renal & mild to moderate hepatic dysfunction. 2nd & 3rd trimesters of pregnancy. Not to be used during lactation.
Adverse Reactions
Headache; abdominal distention & pain, nausea; uterine spasm.
Drug Interactions
Increased elimination rate & decreased bioavailability w/ CYP450 3A4 inducers eg, rifampicin, carbamazepine or St. John's wort (Hypericum perforatum)-containing herbal products. Decreased elimination rate & increased bioavailability w/ CYP450 3A4 inhibitors eg, ketoconazole. Altered release & absorption w/ other vag products eg, antifungals.
