Xalacom

Per mL Latanoprost 50 mcg, timolol 5 mg

Reduction of IOP in patients w/ open-angle glaucoma & ocular HTN who are insufficiently responsive to timolol &/or latanoprost.

1 eye drop in the affected eye(s) once daily in the morning.

Hypersensitivity. Bronchial asthma or a history of bronchial asthma, severe COPD, sinus bradycardia, sick sinus syndrome, SA block, 2nd or 3rd degree AV block not controlled w/ pacemaker, overt cardiac failure, cardiogenic shock.

May cause discoloration of soft contact lenses & eye irritation. Remove contact lens before instillation & reinsert after 15 min. Close monitoring is required w/ frequent or prolonged use in dry eye patients, or in conditions where cornea is compromised. Do not wear contact lens if eyes are red. Not for the treatment of contact lens related irritation. Wait at least 10 min after instilling Xalacom before inserting the contact lens in patients who wear soft contact lenses & whose eyes are not red. Blurring of vision may affect ability to drive or operate machinery. Not to be used during pregnancy & are breastfeeding. Childn & adolescents. Timolol: Closely monitor patients w/ history of severe cardiac disease for signs of cardiac failure. Critically assess hypotension therapy w/ β-blockers in patients w/ CV diseases (eg, CHD, Prinzmetal’s angina & cardiac failure). Patients w/ 1st degree heart block; severe peripheral circulatory disturbance/disorders (eg, severe forms of Raynaud’s disease/syndrome); mild/moderate COPD; corneal diseases. Consider gradual w/drawal of β-adrenergic blocking agents prior to major surgery. May block systemic β-agonist effects. May mask signs & symptoms of hypoglycaemia; hyperthyroidism. Hypersensitivity reactions. Myasthenia gravis. Latanoprost: May gradually increase the brown pigment of the iris, change eyelashes & vellus hair in the treated eye. Treatment may be stopped if increased iris pigmentation ensues. Unilateral treatment can result in permanent heterochromia. Eyelid skin darkening. Macular oedema. Glaucoma. Patients w/ history of herpetic keratitis. Avoid in cases of active herpes simplex keratitis.

Headache; corneal disorder, keratitis, conjunctivitis, blepharitis, eye pain, irritation, hyperaemia, iris hyperpigmentation; HTN. Infection, sinusitis, upper resp tract infection; DM, hypercholesterolemia; depression; cataract, visual field defect; arthritis, vomiting, nausea.

Concomitant use of 2 or more β-adrenergic blocking agents, prostaglandins, prostaglandin analogs or prostaglandin derivatives. Mydriasis occurs when given w/ epinephrine. Potential for additive effects resulting in hypotension &/or marked bradycardia w/ oral Ca-channel blockers, guanethidine or β-blocking agents, antiarrhythmics, digitalis glycosides & parasympathomimetics. Potentiated systemic β blockade (eg, decreased heart rate, depression) w/ CYP2D6 inhibitors (eg, quinidine, fluoxetine, paroxetine) & timolol. The hypertensive reaction to sudden w/drawal of clonidine can be potentiated when taking β-blockers. β-blockers may increase hypoglycemic effect of antidiabetic agents & mask signs/symptoms of hypoglycaemia.
