Levofloxacin hemihydrate
Indications/Uses
Acute maxillary sinusitis, acute exacerbation of chronic bronchitis, pneumonia, skin & soft tissue infections, UTI, acute pyelonephritis.
Dosage/Direction for Use
250-500 mg once daily for 7-14 days. Renal impairment w/ CrCl 20-49 mL/min Initially 500 mg, followed by 250 mg/24 hr; CrCl 10-19 mL/min & haemodialysis patient (chronic ambulatory peritoneal dialysis) Initially 500 mg, followed by 250 mg/48 hr; CrCl 50-80 mL/min No need to adjust dose.
Administration
Should be taken on an empty stomach: Take 1 hr before or 2 hr after meals. Ensure adequate fluid intake.
Contraindications
Hypersensitivity to levofloxacin or other quinolone derivatives. Pregnancy & lactation. Childn <18 yr.
Special Precautions
Patients w/ renal dysfunction, sensitivity to nerve disturbance. Long-term use may cause overgrowth of nonsusceptible microorganisms. Periodic exam for liver, renal & hematopoietic function should be done. Monitor blood glucose level when concomitantly used w/ antihypoglycaemic agents. Elderly may require lower dose.
Adverse Reactions
Headache, drowsiness, insomnia, sleep disturbances, anxiety, depression, hallucination, psychotic reactions, dyspepsia, tremor, taste perversion, nausea, vomiting, diarrhoea, constipation, flatulence, abdominal pain, anorexia, genital moniliasis, leucorrhea, vaginitis, skin rash, urticaria, itching, edema, increased sweating, malaise, fatigue.
Drug Interactions
Absorption may be reduced by Al- & Mg-containing antacids, sucralfate, metal cation, multivit. Oral antidiabetic agents. NSAIDs, theophylline.
