Domperidone
Indications/Uses
Acute nausea & vomiting. Nausea & vomiting due to levodopa & bromocriptine treatment lasting >12 wk. Short-term therapy of functional dyspepsia, nausea & vomiting due to chemo- or radiotherapy in childn.
Dosage/Direction for Use
Functional dyspepsia Adult 10-20 mg 3 times daily & at night. Max duration of treatment: 12 wk. Nausea & vomiting (including those caused by levodopa & bromocriptine) Adult & elderly 10-20 mg every 4-8 hr. Nausea & vomiting due to chemo- or radiotherapy Childn 0.2-0.4 mg/kg/day at 4-8 hr intervals.
Administration
Should be taken on an empty stomach: Take 15-30 min before meals.
Contraindications
Prolactinoma. Whenever stimulation of GI motility might be dangerous. Intolerance to domperidone.
Special Precautions
Long-term treatment. Hepatic or renal dysfunction. Pregnancy & lactation. Infant <1 yr.
Adverse Reactions
Rarely, sedation, extrapyramidal reactions, galactorrhea, gynecomastia, dry mouth, headache, diarrhea, rash, thirst, anxiety, itch. May induce plasma prolactin level.
Drug Interactions
Action inhibited by antimuscarinic agents & opioid analgesics. Decreases the effect of bromocriptine. Decreased bioavailability w/ antacids.
