Why do I need this medicine?
Sotorasib is used to treat a certain type of lung cancer that has spread to other parts of the body in individuals who have failed previous therapy and whose tumour has KRAS G12C (an abnormal change in the gene called KRAS).

This medicine reduces the size of the tumour by stopping cancer cells from growing.
How do I take this medicine?
Take Sotorasib exactly as directed by your doctor or according to the instructions on the label. Do not take more or less than instructed by your doctor.

You may take this medicine with or without food. Try to take it at the same time each day.

Swallow the tablet whole with a glass of water. Do not chew, crush, or split the tablet.

If you have difficulty swallowing the tablet(s), disperse the tablet in 120 mL of non-carbonated, room-temperature water without crushing the tablet. Do not use any other liquids, including acidic beverages (e.g. fruit juices). Stir or swirl the glass gently until the tablet is dispersed into small pieces. Take note that the tablet will not completely dissolve. Drink the mixture immediately or within 2 hours of dispersion. Do not chew the tablet pieces. To make sure that you have taken the full dose, rinse the glass with an additional 120 mL of water and drink it immediately.

If you vomit after taking a dose of Sotorasib, do not take an additional dose. Take the next dose at your regular scheduled time.

The dose of this medicine will be decided by your doctor. Your doctor will advise you on the course of treatment depending on your response to the medication.

Sotorasib must be taken regularly for it to be effective. Continue taking this medicine even when you feel better. Do not stop taking it unless instructed by the doctor.
What should I do if I have forgotten to take this medicine?
If you miss a dose and less than 6 hours have passed, take the missed dose and return to your normal dosing schedule.

If more than 6 hours have passed from your regular scheduled time, skip the missed dose and return to your normal dosing schedule.

DO NOT double a dose under any circumstances.

If you often forget to take your medicine, inform your doctor and pharmacist.
When should I not use this medicine?
Do not breastfeed while you are being treated with Sotorasib and for 7 days after the last dose.
What should I take note of while taking this medicine?
Inform your doctor if you have the following conditions:
  • other lung or breathing problems
  • liver problems
Let your doctor know if you are pregnant.

It is important that you do not get pregnant while taking this medicine. You must use proven birth control methods during Sotorasib therapy and for 7 days after stopping the treatment. Birth control medicines containing hormones may not be effective. You may wish to discuss other reliable methods of birth control with your doctor.

Why is it important to keep my appointments with the doctor?

Keep your appointments with your doctor. Your doctor needs to monitor your condition and check your response to the medication regularly.
  • Certain tests to check the presence of KRAS G12C in the tumour must be done before treatment to know if this medicine is suited for you to take.
  • Blood tests may be performed to make sure that your liver function is normal before you take Sotorasib and thereafter depending on your doctor’s advice.
  • Regular monitoring for signs of lung problems may also be needed.
What side effects could I experience?
Sotorasib may cause any of the following side effects: headache, nausea, vomiting, diarrhoea, stomach pain, constipation, and muscle, joint or back pain.

Some side effects may need immediate medical help. Alert your doctor quickly if you experience any of the following:
  • yellowing of the skin or eyes, abdominal pain, dark coloured urine, tiredness, swelling in the legs and ankles
  • new or worsening shortness of breath, cough, fever.
Inform your doctor if any of these side effects do not go away or are severe, or if you experience other side effects.
Can I take this with other medicines?
Inform your doctor and pharmacist if you are taking any of these medicines:
  • other medicines for cancer e.g. topotecan, enzalutamide, mitotane
  • medicines for epilepsy (fits or seizures) e.g. phenytoin, carbamazepine, phenobarbital
  • medicine that reduces stomach acid production e.g. omeprazole, pantoprazole, famotidine
  • medicines used in organ transplants or certain immune disorders e.g. ciclosporin, tacrolimus, sirolimus
  • medicines for irregular heartbeat e.g. quinidine, digoxin
  • rifampicin (medicine to treat TB, an infection known as tuberculosis)
  • rosuvastatin (cholesterol-lowering medicine)
  • midazolam (medicine for sleep disorders)
  • birth control pills
  • St. John's wort (herbal medicine)
This list does not include all medicines that may interact with Sotorasib.

This medicine should not be taken with antacids (medicines that neutralise stomach acids) as antacids could reduce the effectiveness of Sotorasib. If you must take antacids, take this medicine at least 4 hours before or 10 hours after you have taken the antacid.

Always notify your doctor and pharmacist if you are taking any other medicines, including herbal tonics such as traditional Chinese medicines, supplements, and medicines that you buy without a prescription.
What special dietary instructions should I follow?
Avoid alcohol.
How should I store this medicine?
Store in a cool, dry place away from the reach of children.

Medicines must not be used past the expiry date.

As Sotorasib is a cancer medicine, always return any unused or expired tablets to the clinic, hospital, or pharmacy for disposal. Do not throw it away in the household waste.
