CHD & hypercholesterolemia: Reduces risk of death due to CHD, reduce risk of nonfatal MI, reduces risk of patients who were undergoing myocardial revascularization procedure. Hypercholesterolemia: Reduces total cholesterol & LDL-C in patients w/ primary hypercholesterolemia (type IIa & IIb).
Dosage/Direction for Use
Initially 5-10 mg daily as a single dose, in the evening. Mild to moderate hypercholesterolemia 5 mg daily. Adjust dose w/ interval at least 4 wk, max of 40 mg daily.
Administration
May be taken with or without food: Avoid excessive consumption (>1 L/day) of grapefruit juice.
Contraindications
Active liver disease or persistent increase of serum transaminase. Pregnancy & lactation.
Special Precautions
Perform LFT before treatment & on 6 & 12 wk after the 1st treatment, then following periodically. Alcoholism &/or have a history of liver disease; acute & severe myopathy or secondary kidney failure; not for homozygous familial hypercholesterolemia; type I, IV, & V hypertriglyceridemia. Childn & adolescent.
Adverse Reactions
Abdominal pain, constipation, flatulence, headache, myopathy, rhabdomiolysis, angioneurotic edema, dysfunction of certain cranial nerves, tremor, dizziness, vertigo, memory loss, paresthesia, peripheral neuropathy, paralysis of peripheral nerves; anaphylaxis, angioedema, thrombocytopenia, leucopenia, hemolytic anemia; anorexia, vomiting; alopecia, pruritus; gynecomastia, loss of libido, erectile dysfunction; accelerated cataract, ophthalmoplegia.
