Indications/Uses
Symptomatic relief of acute & recurrent diabetic gastroparesis. Treatment of drug-related post-op nausea & vomiting. Symptomatic short-term treatment of heartburn secondary to reflux oesophagitis.
Dosage/Direction for Use
Tab Adult 1 tab 3 times daily. Syr Adult 10 mL 3 times daily. Syr/Oral drops Childn 1-6 yr 0.1 mg/kg 2-3 times daily, <1 yr 0.1 mg/kg twice daily. Renal impairment Reduce dose to min 60% of normal dose. IV inj Delayed gastric emptying Adult 10 mg over 1-2 min. Childn 6-14 yr 2.5-5 mg, <6 yr 0.1 mg/kg. Prevention of nausea & vomiting associated w/ chemotherapy 1-2 mg/kg over a period of not <15 min, given 30 min prior to chemotherapy. Repeat every 2 hr for 2 doses, then every 3 hr for 3 doses & not >10 mg/kg in each 24 hr. Prevention of post-op nausea & vomiting 10-20 mg IM, given near the end of surgery over 1-2 min period.
Administration
Should be taken on an empty stomach: Take 30 min before meals or at bedtime.
Contraindications
Hypersensitivity or intolerance. Not to be used in epileptic or patients receiving other drugs which are likely to cause extrapyramidal reactions. Obstruction or perforation in the presence of GI hemorrhage. Pheochromocytoma.
Special Precautions
Concomitant use w/ other drugs that can also cause extrapyramidal reactions, eg, phenothiazines. May affect ability to drive or operate machinery. Renal failure. Tab/Syr/Inj: Not to be used during the 1st trimester of pregnancy. Nursing mothers. Tab: Not recommended to be used concomitantly w/ anticholinergic drugs.
Adverse Reactions
Drowsiness, fatigue, lassitude, restlessness, constipation, diarrhea, urticaria, dryness of the mouth, glossal or periorbital oedema, methaemoglobinemia, extrapyramidal reactions eg, parkinsonian symptoms, tardive dyskinesia in childn & elderly.
Drug Interactions
Syr/Oral drops/Inj: Effects of anticholinergic drugs & narcotic analgesics are antagonized by metoclopramide. Additive sedative effects w/ alcohol, sedatives, hypnotics, narcotics or tranquilizers. Increase absorption rate of acetaminophen/paracetamol, tetracycline, levodopa, alcohol/ethanol, cyclosporine, & decrease absorption rate of digoxin. MAOI therapy.
