Vincristine sulfate
Indications/Uses
Neoplastic diseases (combination therapy): Acute lymphocytic leukemia, Hodgkin's disease, non-Hodgkin's lymphomas, small cell lung cancer, breast cancer, cervical cancer, multtiple myeloma, rhabdomyosarcoma, neuroblastoma, Wilm's tumour, paed germ cell tumours, Ewing's sarcoma, osteosarcoma. Monotherapy for idiopathic thrombocytopenic purpura refractory to usual treatment.
Dosage/Direction for Use
IV Adult Usual Dose: 0.4-1.4 mg/m2. Childn Usual Dose: 1.5-2 mg/m2.
Contraindications
Demyelinating form of Charcot Marie-Tooth syndrome. Patients receiving radiation therapy through parts that include the liver.
Special Precautions
Leucopenia, acute uric acid nephropathy, hepatic impairment, jaundice, neurological disease. May cause corneal ulceration on contamination of the eyes. Pregnancy & lactation. Elderly.
Adverse Reactions
Peripheral neuropathy. Loss of deep tendon reflexes & peripheral paraesthesias especially numbness, pain & tingling. Cranial nerve palsy, atrophy, cramps, ataxia, a slapping gait, foot drop & difficulty in walking or inability to walk. Constipation & paralytic ileus, associated w/ abdominal cramps. Urinary tract disturbances, orthostatic hypotension, defective sweating & myoclonic jerks. Depression, agitation, insomnia & hallucination, convulsions & comas. Alopecia. Nausea, vomiting, diarrhea, stomatitis & oral ulceration. Fever & wt loss.
Drug Interactions
Allopurinol may increase the incidence of cytotoxic-induced bone-marrow depression. Increases the cellular uptake of methotrexate.
