Pregabalin
Indications/Uses
Neuropathic pain in adults. Adjunctive therapy in adults w/ partial seizures w/ or w/o secondary generalization.
Dosage/Direction for Use
Dose range: 150-600 mg daily given in 2 or 3 divided doses. Neuropathic pain Recommended starting dose: 75 mg twice daily. May be increased to 150 mg twice daily after 3-7 days & if needed, to a max dose of 300 mg twice daily after an additional wk. Epilepsy Recommended starting dose: 75 mg twice daily. May be increased to 150 mg twice daily after 1 wk & if needed, to a max dose of 300 mg twice daily after an additional wk. Patient w/ CrCl ≥60 mL/min Starting dose: 150 mg twice daily. Max: 600 mg twice daily, 30-60 mL/min Starting dose: 75 mg once or twice daily. Max: 300 mg once or twice daily, 15-30 mL/min Starting dose: 25-30 mg once or twice daily. Max: 150 mg once or twice daily, <15 mL/min Starting dose: 25 mg once daily. Max: 75 mg once daily. Supplementary dose following hemodialysis: Initially 25 mg single dose. Max: 100 mg single dose.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity.
Special Precautions
Rare hereditary problems of galactose intolerance, the Lapp lactase deficiency or glucose-galactose malabsorption. Some diabetic patients who gain wt may need to adjust hypoglycemic medications. May increase occurrence of accidental injury (fall) in the elderly due to dizziness & somnolence. May impair ability to drive & use machines. Effective contraception may be used in women of childbearing potential. Pregnancy & lactation. Childn & adolescents 12-17 yr. Elderly.
Adverse Reactions
Dizziness, somnolence. Increased appetite; euphoric mood, confusion, decreased libido, irritability; ataxia, disturbance in attention, abnormal coordination, memory impairment, tremor, dysarthria, paraesthesia; blurred vision, diplopia; vertigo; dry mouth, constipation, vomiting, flatulence; erectile dysfunction; fatigue, peripheral edema, feeling drunk, edema, abnormal gait; increased wt.
Drug Interactions
Additive impairment of cognitive & gross motor function caused by oxycodone. May potentiate effect of ethanol & lorazepam.
