Indications/Uses
Gastric antisecretory when oral route is not possible eg, GERD in patients w/ esophagitis &/or severe symptoms of reflux, healing of gastric ulcer associated w/ NSAID therapy. Short-term maintenance of hemostasis & prevention of rebleeding in patients following therapeutic endoscopy for acute bleeding gastric or duodenal ulcers.
Dosage/Direction for Use
Gastric antisecretory 20-40 mg IV once daily. Reflux esophagitis 40 mg once daily. Symptomatic treatment for reflux disease 20 mg once daily. Healing of gastric ulcer associated w/ NSAID therapy 20 mg once daily. Short-term maintenance of hemostasis & prevention of rebleeding of gastric & duodenal ulcers 80 mg bolus infusion over 30 min, followed by continuous IV infusion of 8 mg/hr given over 3 days (72 hr).
Contraindications
Hypersensitivity. Concomitant use w/ atazanavir & nelfinavir.
Special Precautions
Exclude gastric malignancy before treatment. Pregnancy & lactation.
Adverse Reactions
Leucopenia, thrombocytopenia, agranulocytosis, pancytopenia, hypersensitivity reactions, peripheral oedema, hyponatremia, insomnia, agitation, confusion, depression, aggressiveness, hallucination, headache, dizziness, paraesthesia, somnolence, taste disturbances, blurred vision, vertigo, bronchospasm, abdominal pain, constipation, diarrhea, flatulence, nausea/vomiting, dry mouth, stomatitis, GI candidiasis, increased liver enzyme, hepatitis (w/ or w/o jaundice), liver damage, encephalopathy in patients w/ liver disease, local reactions at inj site, dermatitis, pruritus, skin redness, urticaria, alopecia, photosensitivity, erythema multiforme, SJS, TEN, arthralgia, myalgia, muscle weakness.
Drug Interactions
Ketoconazole, diazepam, citalopram, phenytoin, warfarin, other coumarin derivatives, cisapride, atazanavir, nelfinavir, saquinavir, voriconazole.
