Mycophenolate mofetil
Indications/Uses
In combination w/ ciclosporin & corticosteroids for immunosuppression to prevent rejection of transplanted kidney, heart or liver.
Dosage/Direction for Use
Adult Kidney transplant 4 tab daily taken as 2 separate doses (2 in the morning, 2 in the evening). Take 1st dose w/in 72 hr after transplant operation. Heart transplant 6 tab daily taken as 2 separate doses (3 in the morning, 3 in the evening). Take 1st dose w/in 5 days following transplant operation. Liver transplant 6 tab daily taken as 2 separate doses (3 in the morning, 3 in the evening). Take 1st dose at least 4 days after transplant operation & when able to swallow oral medicines. Childn 2-18 yr Kidney transplant 600 mg/m2 bd.
Administration
May be taken with or without food: Swallow whole, do not break/crush.
Contraindications
Hypersensitivity to mycophenolate mofetil or mycophenolic acid. Women of childbearing potential not using highly effective contraception. Pregnancy & lactation.
Special Precautions
Patients w/ evidence of infection, unexpected bruising &/or bleeding; existing or history of any digestive system problems. Increased risk of skin cancer. Reports of hypogammaglobulinaemia in association w/ recurrent infections in combination w/ other immunosuppressants; bronchiectasis in adults & childn in combination w/ other immunosuppressants. Do not donate blood during therapy & for at least 6 wk following treatment discontinuation. Increased risk of spontaneous abortion & congenital malformations in case of exposure during pregnancy. Women of childbearing potential should simultaneously use 2 reliable forms of contraception before starting & during therapy, & for 6 wk after treatment discontinuation. Sexually active (including vasectomized) male patients & their female partners should use highly effective contraception during treatment & for at least 90 days after treatment cessation. Men should not donate semen during therapy & for 90 days following treatment discontinuation. No available data to recommend use in childn 2-18 yr who have received heart &/or liver transplant.
Adverse Reactions
Diarrhoea, decreased WBCs &/or RBCs, infection, vomiting.
Drug Interactions
Potential interaction w/ azathioprine or other immunosuppressants, cholestyramine, rifampicin, antacids, phosphate binders; live vaccines.
