Rosuvastatin
Indications/Uses
Reduces LDL-C & increases HDL-C when diet & exercise are inadequate to correct cholesterol levels in patients w/ high cholesterol level &/or at risk for heart attack or stroke due to atherosclerosis.
Dosage/Direction for Use
Reduce cholesterol levels Initially 10 mg, then increase to 20-40 mg if necessary; 4-wk gap between every dose adjustment. Max daily dose: 40 mg. Reduce risk of heart attack, stroke or related health problems 20 mg daily.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Concomitant use of ciclosporin. Patients w/ repeated or unexplained muscle aches or pains; liver disease; severe kidney problems. Pregnancy & lactation. 40 mg (highest dose): Regular consumption of large amounts of alcohol. Concomitant use of fibrates (eg, gemfibrozil, fenofibrate). Patients w/ impaired thyroid gland; history of repeated or unexplained muscle aches or pains, personal or family history of muscle problems or previous history of muscle problems when taking other cholesterol-lowering medicines; moderate kidney problems. Patients of Asian origin.
Special Precautions
Continue w/ cholesterol-lowering diet & exercise during therapy. Caution before treatment initiation in case of kidney or liver problems; history of repeated or unexplained muscle aches or pains, personal or family history of muscle problems or previous history of muscle problems when taking other cholesterol-lowering medicines; regular consumption of large amounts of alcohol; impaired thyroid gland; concomitant use w/ fibrates; concomitant use w/ HIV drugs eg, lopinavir/ritonavir; elderly >70 yr; severe resp failure; Asian origin; concomitant use w/ fusidic acid or use w/in the last 7 days. Perform liver function test before & during treatment. Increases in HbA1c & fasting serum glucose levels; caution in diabetic patients or patients at risk of developing DM. Rare post-marketing reports of cognitive impairment associated w/ statin use. Patients intolerant to some sugars (lactose or milk sugar). Concomitant use w/ PIs is not recommended. Should not be given in childn <10 yr.
Adverse Reactions
Headache, stomach pain, constipation, feeling sick, muscle pain, feeling weak, dizziness, proteinuria, diabetes.
Drug Interactions
Potential interaction w/ ciclosporin, warfarin, fibrates (eg, gemfibrozil, fenofibrate) or any cholesterol-lowering drugs (eg, ezetimibe), indigestion remedies, erythromycin, OCs, hormone replacement therapy, lopinavir/ritonavir. Risk of rhabdomyolysis w/ fusidic acid.
