Donepezil HCl
Indications/Uses
Symptoms of dementia (eg, increasing memory loss, confusion & behavioural changes) in adults diagnosed as having mild & moderately severe Alzheimer's disease.
Dosage/Direction for Use
Initially 5 mg every night, subsequently increased to 10 mg every night after 1 mth. Max: 10 mg every night.
Administration
May be taken with or without food: Take at bedtime. Place tab on the tongue & allow to disintegrate before swallowing w/ or w/o water.
Contraindications
Hypersensitivity to donepezil HCl, or to piperidine derivatives.
Special Precautions
Patients w/ stomach or duodenal ulcers; seizures or convulsions; irregular or very slow heart beat; asthma or other long term lung disease; difficulty passing urine. Discontinue if patient develops neuroleptic malignant syndrome; markedly elevated blood creatine phosphokinase (CPK) &/or symptoms indicative of rhabdomyolysis. Use w/ caution in patients w/ prior history of muscular disorders, uncontrolled hypothyroidism. Concomitant use w/ medications causing rhabdomyolysis (eg, statins, antipsychotics, SSRIs/SNRIs). May impair ability to drive or operate machinery. Liver problems or hepatitis. Mild kidney disease. Pregnancy & lactation. Childn & adolescents <18 yr.
Adverse Reactions
Diarrhoea, feeling sick, headaches. Muscle cramp, tiredness, insomnia, common cold, loss of appetite, hallucinations, unusual dreams including nightmares, agitation, aggressive behaviour, fainting, dizziness, stomach feeling uncomfortable, rash, itching, passing urine uncontrollably, pain, accidents (eg, falls, accidental injury).
Drug Interactions
Other Alzheimer's disease medicines (eg, galantamine); pain killers or treatment for arthritis (eg, aspirin, NSAIDs [eg, ibuprofen or diclonefac Na]); anticholinergics (eg, tolterodine); antibiotics (eg, erythromycin, rifampicin); antifungals (eg, ketoconazole); anti-depressants (eg, fluoxetine); anticonvulsants (eg, phenytoin, carbamazepine); medicines for heart conditions (eg, quinidine, β-blockers [eg, propranolol & atenolol]); muscle relaxants (eg, diazepam, succinylcholine); general anaesth; medicines obtained w/o prescription (eg, herbal remedies); alcohol.
