Atorvastatin Ca
Indications/Uses
Lowers blood cholesterol & triglycerides in adults & childn ≥10 yr when a low fat diet & other non-medicinal treatments eg, exercise & lifestyle changes have failed.
Dosage/Direction for Use
Adult & childn ≥10 yr 10 mg once daily.
Administration
May be taken with or without food: Avoid excessive consumption (>1 L/day) of grapefruit juice.
Contraindications
Hypersensitivity. Liver disease; abnormal blood tests for liver function. Excessive alcohol intake. Intolerance to some sugars. Woman of childbearing potential not using adequate contraceptive measures. Pregnancy & lactation.
Special Precautions
Kidney problems, hypothyroidism, any muscle disorders (affecting either the patient or other family members), previous muscular problems during treatment w/ other lipid-lowering medicines, history of heavy alcohol consumption. Perform blood tests to determine risk of muscle-related side effects. Perform liver function tests before therapy initiation & thereafter when clinically indicated. Stop taking Divator about 1 mth before planning to conceive. Elderly >70 yr.
Adverse Reactions
Nausea, abdominal pain, constipation, wind, indigestion, headache, muscle pain, weakness, diarrhoea & insomnia.
Drug Interactions
Potential interaction w/ immunomodulating agents eg, cyclosporin; antihistamines eg, terfenadine, astemizole; macrolide antibiotics eg, erythromycin; azole antifungal eg, ketoconazole, itraconazole; rifampicin; medicines to regulate lipid levels eg, fibric acid derivatives, niacin, colestipol; some Ca channel blockers eg, nifedipine; some benzodiazepines eg, diazepam; PIs eg, nelfinavir; digoxin, warfarin, OCs, antacids, phenytoin. Avoid co-administration of strong CYP3A4 inhibitors (eg, ciclosporin, telithromycin, clarithromycin, delavirdine, stiripentol, ketoconazole, voriconazole, itraconazole, posaconazole & HIV PIs including ritonavir, lopinavir, atazanavir, indinavir, darunavir). Avoid concomitant use in patients taking telaprevir. Do not exceed 40 mg daily when taking w/ boceprevir. Do not drink >1-2 small glasses of grapefruit juice per day.
